Studies have shown that drug use can alter the mother-infant relationship and, consequently, the development of the offspring. Given this, the present study aimed to evaluate whether the administration of the substance 25H-NBOMe (serotonergic receptor agonist) would affect the MC and the consequences for the offspring. Females were treated with different doses of 25H-NBOMe (0.3mg/kg and 1.0mg/kg). The substance was applied to lactating females on the fifth postpartum day (D5) and non-lactating females during the diestrus phase. HTR and Open Field tests were performed to evaluate the drug's psychedelic activity. In lactating females, the tests for psychedelic evaluation were performed on D5 together with the evaluation of the MB, called motivated (MMB). Throughout the lactation period, the MB was evaluated, called continuous (CMB). The measurement of the weight gain of lactating females occurred from the day of mating until weaning. In the offspring, the registration occurred from the 2nd to the 32nd postpartum day. With the litter of lactating females, tests of Playing Behavior and Social Interaction were performed. Data were evaluated using the Graph Pad Prism version 8.0 program. Thus, our results suggest that the effect of 25H-NBOMe on CMB, despite changing some parameters, there is no statistical difference between treatment and control groups. On the other hand, in the MMB, we found changes in the parameters of the recovery of the pups and an increase in latency. There was no difference between the groups in the weight gain assessment of lactating females and litter. Both lactating and non-lactating females make twitching movements of the head, but there is no change in the open field test. Finally, in the litter's behavioral tests, there is a change in some parameters of playing behavior but not in the social interaction of the offspring.
