This work intended to evaluate cytotoxicity and inflammation response of the new material for pulpotomy - Bio-C Pulpo (Angelus®). During the in vitro study, an MTT test has been done to L929 fibroblast cells culture in order to analyze cellular viability. For the in vivo study, 30 male rats (Rattus norvegicus, albinus, Wistar) have been used. The animals were divided in 5 different periods - 7, 15, 30, 60 and 90 days, thus 6 animals per period, in which 3 implants (poliethilenl pipes) containing the following material: 1) Bio- C Pulpo (Angelus®), (2) white MTA (Angelus®), (3) Control – empty pipe, were inserted into their dorsal region. The inflammatory cells have been analyzed morphometrically as so as the imuno-histochemic technique for TNFα AND IL-1β. The results showed that the viability wasn’t affected by non diluted extracts and in ½, ¼ and ⅛ solutions, MTA and Bio-C Pulpo have showed more viability than the Control. Bio-C Pulpo has presented cellular viability similar to MTA’s and in both materials the best response was in 1/1 solution in 24 hours and in 48 hours Bio-C reached best response than MTA. By evaluating inflammatory response of the material in different periods, the MTA group presented more intense inflammatory response in 7, 15 and 30 days than in 60 and 90 days (p<0,05). In Bio-C Pulpo case, it has been verified a bigger number of inflammatory cells in 7 and 15 days than the other periods (p<0,05). In 7 days the Control presented the biggest inflammatory cells (p<0,05). When analyzing different material in the same period, Bio-C Pulpo had the biggest inflammatory response (p<0,05). In 60 days, Bio-C Pulpo and MTA presented more inflammatory cells than Control (p>0,05). In 90 days, all the materials showed similar inflammatory response (p>0,05). The TNFα and IL1β pro inflammatory citocinese imuno-histochemic analysis allows to say that both materials presented inflammatory response reduced in 7, 15 and 30 days, moderated response in 60 and 90 days and analyzing citocinese in the same period the materials and the control were similar statistically.
