These microorganisms are sources of secondary metabolites, which are the focus of study in natural product science due to their biological potentials. The present study performed a systematic review on endophytic fungi of the genus Aspergillus sp., which presented a large number of metabolites (230) with anti-inflammatory properties described in the literature. Among the compounds observed, four showed multitarget inhibition of the main pro-inflammatory pathways. A database of anti-inflammatory molecules from the genus was developed and a QSAR machine learning model to predict nitric oxide (NO) inhibition of the inflammatory pathway was created. Leaves of the species Poincianella pluviosa (Sibipiruna) were collected and extracts and fractions of endophytic fungi, were evaluated for anti- inflammatory (ex vivo) and antiparasitic (in vivo) activity . Extracts FPD4 and FPD13 showed moderate anti-inflammatory activity by inhibiting PGE2 by 87.25% and 77.72%, respectively. FPD4 and FPD13 extracts were analyzed in HPLC-MS, and compound annotation was possible by MS1 and MS2 spectra. A total of 81substances were dereplicated using internal database and 3 substances (Nigrosporolide, tirosol and decarestrictin A) were isolated and characterized by spectroscopic (NMR) and spectrometric (UHPLC-MS/MS) techniques. Analyses performed with crude extract FPD13 showed that concentrations of 4 and 2 mg/mL of the extract, inhibited the larval hatching of Haemonchus contortus eggs by 100% .The strain of FPD13 was isolated and classified as Nigrospora. Zirmmini by morphological characteristics and sequencing of the ITS (Internal Transcribed Spacer) region with phylogenetic analysis (GenBanK reference sequences). A classical review on the monophyletic genus Nigrospora demonstrated the existence of 271 bioactive metabolites described in the literature, belonging to 21 different species/cultures of the genus. In addition, polyketides, alkaloids and butenolides have been shown to be the main classes of natural products described in endophytic fungi.
