The drugs used in conventional treatments are generally toxic and often induce to tumor cells resistance. Thus, it is extremely important to identify compounds with antitumor activity that can improve the therapeutic approaches against breast cancer. 7-epiclusianone is a tetraprenyladed benzophenone isolated from the hexane extract of Garcinia brasiliensis. Studies have shown that 7-epiclusianone has different biological properties, which include antimicrobial, anti-spasmodic, leishmanicide, anti-inflammatory and analgesic activities. Futhermore, a previous study showed that this compound inhibits the proliferation rate of cell lines derived from human cancers. Therefore, the aim of this study was to analyze the cytotoxic and/or antiproliferative effects of 7-epiclusianone on human breast carcinoma cell lines. Results showed that 7-epiclusianone reduces the cell viability of MCF-7 and Hs 578T lines in a time and concentration dependent manner. Antiproliferative activity of this compound on studied cell lines are relates to its ability to inducing cell cycle arrest in G1/S transition, while the cytotoxic activity was attributed, at least in part, to its pro-apoptotic potential. 7-epiclusianone reduced the mRNA levels of both cyclins D1 and E in MCF-7, but only cyclin E expression was downregulated in Hs 578T.
