Dissertação (Mestrado em Ciências Farmacêuticas) - Universidade Federal de Alfenas, Alfenas, MG, 2011 .https://repositorio.unifal-mg.edu.br/handle/123456789/278Tea made from the stem bark of Byrsonima intermedia A. Juss. is popularly used to treat diarrhea and dysentery. Their shells contain polyphenols which can be attributed to antimicrobial, anti-hemorrhagic, anti-diarrheal and anti-inflammatory. In this study the extract lyophilized bark of Byrsonima intermedia was characterized by chemical tests to assess the presence of secondary metabolites, quantification of phenolic and flavonoid content and chromatographic profiles by high performance liquid chromatography (HPLC) were evaluated antinociceptive, anti-inflammatory and antidiarrheal effects lyophilized extract of bark of Byrsonima intermedia (ELBi) to make their use safer as a medicinal plant. The formalin test, hot plate and von Frey filaments were used to evaluate the antinociceptive activity in mice. To evaluate the anti-inflammatory tests were conducted granuloma, peritonitis induced by LPS (lipopolysaccharide) and paw edema induced by carrageenin in rats. To evaluate the antidiarrheal effect of the extract was tested in vivo, the test of intestinal transit and in vitro, the isolated organ bath. The aqueous extract of bark of Byrsonima intermedia revealed the presence of 37.3 mg gallic acid/g sample and 6.3 mg quercetin/g of sample. In the formalin test the extract showed a significant antinociceptive effect when compared with the control at doses of 100 and 300 mg/kg in the first phase, and three doses 30-300 mg/kg in the second phase. In the hot plate test ELBi at doses of 30-300 mg/kg increased the latency time compared to the control group, as well as morphine significantly. In testing with von Frey filaments was observed a significant decrease in paw withdrawal threshold of the three doses 30-300 mg/kg ELBi, as well as treatment with indomethacin 10 mg/kg. In the evaluation of anti-inflammatory extract at doses of 30, 100 and 300 mg/kg reduced the weight of granuloma in 37.9%, 36.9% and 45.4% respectively in the granuloma test. In the test of peritonitis inhibition values in the recruitment of leukocytes were 21.4, 32.3 and 40.12% for the extract at doses of 30, 100 and 300 mg/kg, respectively. As for the paw edema induced by carrageenin inhibition values were 11.8, 31.6 and 25.2% for doses of 30, 100 and 300 mg/kg of extract, respectively. To evaluate the antidiarrheal effect in the test of intestinal transit was observed that the average length traveled by the charcoal when the animals were pretreated with ELBi at a dose of 300 mg/kg significantly decreased when compared with control. In studies in vitro contractility of the tissue was decreased significantly in front of mediators such as acetylcholine, Ca+2 and K+. With the results we can infer that the aqueous extract of bark of Byrsonima intermedia exerts antinociceptive, anti-inflammatory and antidiarrheal.application/pdfAcesso Abertohttp://creativecommons.org/licenses/by/4.0/MalpighiaceaeAnalgésicosAntiinflamatóriosAntidiarréicosCIENCIAS DA SAUDE::FARMACIAEfeito antinociceptivo, anti-inflamatório e antidiarreico do extrato liofilizado da casca do caule de Byrsonima intermedia A.
